For the analysis,50 ml of each extract was transferred
with a pipette into a 100-ml round-bottom flask and the solvent
was distilled off in vacuum on a rotor evaporator
(Watterbath-480,Büchi,Switzerland) at a water bath temperature
within 60 – 90°C.The dry residue was dissolved in
5 ml of chloroform and this solution was analyzed by TLC
on 10 \x06 10 cm Merck G-60 plates.Three samples were applied
with a micropipette onto the start line (0.07 ml of sample
solution and 0.01 ml of a 25% aqueous ammonium hydroxide
solution) and the plate was dried for 20 min in air.
Then the plate was placed into a TLC chamber (presaturated
for 1 h) with a chloroform – ethanol (90 :10) mixture and
chromatographed in the ascending mode at 25°C.
The bands containing sanguinarine and chelerythrine
(colored orange and yellow,Rf 0.6) were quantitatively
transferred into 50-ml flasks with ground-glass joints and
10 ml of ethanol acidified with 1% H2SO4 added.The solutions
were stirred for 5 min,transferred into centrifuge tubes,
and centrifuged for 20 min.The quantitative analysis for alkaloids
was performed (according to VFS 42-2666–89) by
spectrophotometry at 452 nm (Hitachi EP-3T spectrophotometer).
