If you are talking about plasmid in bacterial strain,then just grow it on relative antibiotic medium,either plate or liquid.With plate,you can begin with pure culture (from single colony),while with liquid,you can extract more plasmid DNA with it or you can store the strain in freeze (-80C,plus some glycerol:16 to 50%,depending on your preference).
